[Source] This product is the dried mature pulp of Cornus officinalis Sieb. et Zucc., a plant of the Cornaceae family. The fruits are harvested when the skin turns red in late autumn and early winter, baked over low heat or slightly blanched in boiling water, and the core is promptly removed and dried.

[Properties] This product is in irregular flake or capsule shape, 1 to 1.5 cm long and 0.5 to 1 cm wide. The surface is purple-red to purple-black, wrinkled and shiny. Some have round persistent calyx scars at the top and fruit pedicel scars at the base. Soft texture. It has a faint smell and tastes sour, astringent and slightly bitter.

【Identification】 The powder of this product is reddish brown. The epidermal cells of the pericarp are polygonal or rectangular in surface view, with a diameter of 16 to 30 μm, with bead-like thickening of the antibulbar wall, granular keratin thickening of the outer flat wall, and the cell cavity containing light orange-yellow substances. Mesocarp cells orange-brown, mostly wrinkled. There are few calcium oxalate clusters with a diameter of 12 to 32 μm. The stone cells are square, oval or rectangular, with obvious pits and large cell cavities.

[Content Determination] Take about 1g of crude powder of this product, weigh it accurately, put it in a Soxhlet extractor, add appropriate amount of ether, heat and reflux to extract for 4 hours, recover the ether from the extract until dry, soak the residue with petroleum ether (30-60℃) twice, 15ml each time (about immersion for 2 minutes), pour off the petroleum ether, add anhydrous ethanol-chloroform (3:2) mixture to the residue, heat it slightly to dissolve, transfer it to a 5ml volumetric flask, and dilute to the scale, shake well, and use it as the test solution. Take another ursolic acid reference substance and add anhydrous ethanol to prepare a solution containing 0.5 mg per 1 ml as the reference substance solution. According to the thin layer chromatography method (Appendix Ⅵ B) test, 5μl or 10μl of the test solution and 4μl and 8μl of the reference solution are respectively crossed onto the same silica gel G thin layer plate, and cyclohexane-chloroform-ethyl acetate (20:5:8) is used as the developing solvent. After development, the plate is taken out and air-dried, and sprayed with 10% ethanolic sulfuric acid solution. Heat at 110°C for 5 to 7 minutes until purple spots appear. The plate is taken out and covered with a glass plate of the same size on the thin layer plate. Fix the surrounding with tape and scan according to the thin layer chromatography method (Appendix Ⅵ B Thin Layer Scanning Method). The wavelength is: λs=520nm, λR=700nm. The integrated absorbance value of the test sample and the integrated absorbance value of the reference are measured and calculated to obtain the result.
