1. Pharmacodynamics

The antibacterial spectrum is similar to that of penicillin, and it has a strong antibacterial effect on Gram-positive bacteria such as Staphylococcus aureus (including penicillin-resistant strains), Pneumococcus, Streptococcus, Bacillus anthracis, Erysipelothrix rhizopus, Listeria, Putrefactive Clostridium, and Clostridium difficile. Sensitive Gram-negative bacteria include Haemophilus influenzae, meningococci, Brucella, Pasteurella, etc., while insensitive ones are mostly enteric bacilli such as Escherichia coli and Salmonella. also. It also has a good effect on Campylobacter, certain spirochetes, mycoplasmas, rickettsia and chlamydia.

Bacteria have shown increasing resistance to erythromycin, and induced resistance may occur with longer courses of treatment. Bacteria often become resistant to erythromycin due to posttranscriptional methylation of adenine residues on 23S ribosomal RNA. In addition, drug resistance can also occur by preventing drugs from penetrating the bacterial cell membrane.

2. Pharmacokinetics

Erythromycin can be absorbed when taken orally, but it can be destroyed by stomach acid. When pigs were fed erythromycin enteric-coated tablets at a dose of 50 mg per kg of body weight, the peak blood concentration was reached in 1 hour, and 73% of the pigs maintained a concentration greater than 1 μg/mL in the blood for a long time. After intravenous injection of erythromycin lactobionate (8 mg/kg) in pigs, the blood concentration was 7.62 μg/mL 5 minutes later, dropped to 1 μg/mL after 1 hour, and only 0.06 μg/mL remained after 6 hours. The same amount was injected intravenously into cattle once, and the serum concentration was 14.09 μg/mL at 0.08 h, dropped to 2.88 μg/mL at 1 h, and still remained at 0.50 μg/mL at 6 h.

The peak blood concentration (1.3 μg/mL) of laying hens was reached 0.5 h after intramuscular injection of 25 mg/kg and 1 h after oral administration of 100 mg/kg (0.59 μg/mL). The blood concentration was very low (0.08~0.76 μg/mL) when the drug was fed in drinking water (0.5~1g/L).

Erythromycin can be widely distributed in various tissues and body fluids, with an apparent distribution volume of 3.3 L/kg for pigs and 1.62 L/kg for cattle. The highest levels are in the liver and bile and the lowest in cerebrospinal fluid.

A considerable amount of erythromycin is inactivated in the liver and is mainly excreted through bile, part of it is reabsorbed in the intestine, and a small amount is excreted unchanged through urine. The elimination half-life is 1.21h in pigs and 1.97h in cattle.

It is also effective in treating mycoplasma disease (chronic respiratory disease) and infectious rhinitis in chickens. It can also be formulated into eye ointment or cream for skin and eye infections.

It has a good therapeutic effect on fish white head and white mouth disease, gill rot and kidney disease.

Erythromycin can be used as an alternative drug for animals allergic to penicillin.
