Synergistic effect of subthreshold dose of sodium pentobarbital: Liu Guoqing et al. reported that mice were intraperitoneally injected with different doses of Cyperus rotundus volatile oil, 0.03, 0.06 and 0.10 ml/kg (1/10, 1/5, 1/3 median lethal dose, respectively). 30 minutes after administration, mice in each group were intraperitoneally injected with a subthreshold dose of sodium pentobarbital, 20 mg/kg. The disappearance of righting reflex was used as the sleep indicator, and the number of sleeping mice in each group was observed. The results showed that different doses of Cyperus rotundus volatile oil could significantly synergize with the hypnotic effect of sodium pentobarbital on mice (P<0.01).

2. Anesthetic effect on normal rabbits: Rabbits were slowly intravenously injected with different doses of Cyperus rotundus volatile oil, 0.050, 0.075 and 0.100 mg/kg, and the average anesthesia time was 9.0, 15.0 and 28.5 minutes, respectively. After administration, the righting reflex of the animals in each group disappeared rapidly. In the 0.050 mg/kg dose group, the pain response and corneal reflex of the rabbits were sluggish, but the auditory response was present; in the other two dose groups, the pain response and corneal reflex of the rabbits completely disappeared, but the auditory response was present. Rabbits in each group experienced limb stiffness after administration, which disappeared after about 3 minutes.

3. Synergistic scopolamine anesthetic effect: Taking the disappearance of righting reflex as the anesthesia index, the average anesthesia time of rabbits in each group was observed. The first group was given an intravenous injection of 0.075 ml/kg of Cyperus rotundus volatile oil, and all of them showed loss of righting reflex. The second group received intraventricular injection of scopolamine. The third group received an intravenous injection of 0.035 ml/kg of Cyperus rotundus volatile oil (no loss of righting reflex), followed by an intraventricular injection of 2 mg/kg of scopolamine. The results showed that the dose of 0.035 ml/kg had no anesthetic effect, but could significantly prolong the anesthetic time of scopolamine, but did not affect the depth of anesthesia.
